Blocks of ambrosium can be crafted using 9 Ambrosium Shards in a 3x3 crafting grid, with one in each slot.

Altar Enchanting

A Block of Ambrosium can be used as fuel for the Altar, being equivalent to 10 Ambrosium Shards.

Fuel

In addition to their use in an Altar, blocks of ambrosium can be used as furnace fuel; each block lasts 800 seconds (16000 ticks), smelting as many as 80 items.
